About Dan
I began my neuro-oncology career at UCLA in graduate school working to characterize the molecular biology of patient derived brain tumor cell cultures, delineate the mechanism of resistance to mTOR inhibition, and study differential sensitivity to therapy. As you see in my resume I achieved these goals, delineated a novel pathway of resistance to mTOR inhibition, and published all three studies as a first author.
As a post doctoral scholar at the Sloan Kettering Institute, I joined the Luis Parada lab to study cancer stem cells based on their genetically engineered mouse model. First, we characterized their cells of origin, neural stem cells. Through scRNA-Seq and transcriptomics, I discovered a gene signature for glioblastoma stem cells, and moreover, a cell surface marker, F3, which we used to sort out quiescent glioblastoma stem cells and demonstrate their unique self-renewal capacity (co-first author Study in Developmental Cell).
In 2020, I was recruited by Voyager Therapeutics to help start up the neuro-oncology group. I served as a successful program lead of the vHER2 program, established pre-clinical efficacy models, and developed AAV mediated gene therapy with enhanced Fc modified vectorized antibodies to treat HER2+ breast cancer brain metastasis. I designed, developed, and performed in vivo orthotopic xenograft studies for PK/PD and preclinical pharmacology. I performed scRNA-Seq to delineate the innate immune response to therapy. I led the design of mutating the Fc region to obviate FcRN binding and reduce peripheral levels of our therapeutic antibody to have selective exposure in the brain and reduce the risk of cardiotoxicity. As the project lead, I oversaw the vHER2 program including producing timelines and carrying out oversight of CROs for both mouse and NHP studies (Presentation at SNO/ASCO 2023). I have 4 patents published from this work (WO(concealed information), WO(concealed information), WO(concealed information), WO(concealed information)) including the novel vHER2 therapy, a novel class of BBB penetrant AAV, and 14 more pending patents.
